What are peptides?

Peptides are chains of amino acids connected by peptide bonds. Depending on the convention used, shorter chains are generally described as peptides or oligopeptides, while longer chains can be described as polypeptides or proteins. NCBI describes peptides as compounds made from amino acids joined by peptide bonds in linear, branched, or cyclic structures.

The sequence, stereochemistry, terminal modifications, disulfide bonds, and other chemical modifications determine a peptide's molecular identity and properties.

What are biologics?

Biological products are a broad category that includes products such as vaccines, blood components, cells, gene therapies, tissues, and recombinant therapeutic proteins. FDA notes that biologics are often complex molecules or systems whose characterization and manufacturing can be more challenging than conventional small-molecule chemistry.

Not every research peptide is automatically a biologic under a particular regulatory definition. Classification depends on the molecule and applicable law.

What is lyophilization?

Lyophilization, commonly called freeze-drying, removes water from a frozen material under controlled vacuum conditions. The process generally involves freezing, primary drying by sublimation, and secondary drying to reduce residual moisture.

Freeze-drying is widely used in research and biopharmaceutical development because converting a peptide or protein formulation to a dry solid can improve storage stability compared with an aqueous formulation, although the process itself can affect aggregation or molecular conformation.

What is HPLC testing?

High-performance liquid chromatography (HPLC) separates chemical components as they pass through a chromatographic column. For peptide analysis, reverse-phase HPLC with UV detection is commonly used to assess chromatographic purity and generate a chromatogram showing the principal peak and related peaks.

HPLC purity is not the same thing as identity, net peptide content, sterility, endotoxin status, or heavy-metal testing. Those questions require appropriate analytical methods.

How to evaluate a research-lot analytical report.

What a strong COA should identify

  • Exact compound name and, where applicable, CAS number.
  • Manufacturer or submitting party and laboratory identity.
  • Lot/batch number that matches the material being evaluated.
  • Test date and report/accession number.
  • Analytical method(s), not merely a headline purity percentage.
  • Chromatogram or analytical output where appropriate.
  • Identity result, typically supported by mass spectrometry or another validated identity method.
  • Purity result and an explanation of how purity was calculated.
  • Any separately ordered measurements such as net peptide content, endotoxin, residual solvents, heavy metals, or sterility.

How to read the numbers

  • HPLC purity: usually an area-percentage measurement based on the chromatographic method. It does not automatically tell you the mass of peptide in the vial.
  • LC-MS/MS: provides molecular-mass/identity evidence and can detect unexpected peptide species depending on the method and reference library.
  • Net peptide content: attempts to quantify actual peptide material rather than simply reporting chromatographic area percentage.
  • Endotoxin: measures bacterial lipopolysaccharide activity using an appropriate assay such as LAL. It is a separate analytical question from purity.
  • Heavy metals: typically requires an elemental-analysis method such as ICP-MS.
  • Sterility/microbial testing: requires microbiological methods and is not established by HPLC alone.
Important: A COA is evidence from a particular sample or lot under a particular method. A vendor-provided COA should be checked against the lot number and, where possible, independently verified. A high HPLC percentage by itself does not prove identity, concentration, sterility, endotoxin status, or absence of every possible contaminant.
